Opponent attacked my leader, I used Blocker to redirect to a character. They then hit my leader AGAIN the same turn with a second attacker. Can I Blocker again with a different character?

Yes — Blocker is once per character per battle, not once per turn. A different untapped Blocker character can redirect the second attack too.
